- 介紹: N-(2-羥乙基)哌嗪-N’-(2-乙磺酸),是可滿足于所有生物學研究的最佳緩沖液之一。絕大部分生理PHS下HEPES是一種兩性離子有機化學緩沖劑,屬于Good’s緩沖試劑之一,pH 6.0-8.5范圍內的緩沖能力最強。HEPES應用非常廣泛,包括:1)用作細胞培養(yǎng)的緩沖體系,常用工作濃度為10-25 mM;與無機碳酸氫鹽緩沖系統(tǒng)相比,在組織或者器官培養(yǎng)中表現(xiàn)出更好的pH控制能力。不像碳酸氫鹽,HEPES不為細胞供給營養(yǎng)。只有當需要長時間在CO2培養(yǎng)箱內處理細胞,才會加入培養(yǎng)基內以增強緩沖能力。2)用作兩性電解質溶液;HEPES能溶解兩性電解質,制備<1 pH單位寬度的pH梯度以用于等電聚焦電泳實驗;3)用作電鏡觀察蛋白沉積技術合適的緩沖體系,因HEPES不會對金屬基板造成負面影響;4)可用作定量和選擇性測定抗原-抗體反應的緩沖試劑。
